Thin-layer polyacrylamide electrophoresis mal. Proband had markedly high Km and and kinetic studies were used to differenmother intermediately high Km. In the tiate three different mutant erythrocyte third family, proband and mother had pyruvate kinase (PK) in congenital nonfaster migrating PK than the normal. spherocytic hemolytic anemia. In the first Proband had high Km and mother had infamily, proband and father had PK which termediately high Km. The parents in the migrated faster than the normal and second and third families were conhigh Michaelis-Menten constants (Km), sanguineous. Inherited molecular abwhereas mother had normal migration normalities of erythrocyte PK were proven and kinetics, but low PK activity. In the to exist by abnormal electrophoretic mosecond family, proband and mother had bilities, as well as abnormal results of PK which migrated slower than the norkinetic studies.
